The new proposal creates a rapid diagnostic kit, simple and affordable for any health centre

Experts from the University of Cadiz and from the Hospital of Puerta del Mar has patented a process for diagnosing a type of clotting disorder, by identifying a new compound that is deficient in platelets from sick individuals.

The Platelets are blood components that play a key role in coagulation. Some of these processes are regulated through the release of compounds, which are stored in platelets, cellular elements within the so-called "dense granules" or "delta granules.

Thus, “the new method is the study of the levels of one of the compounds we have recently found abundant in the platelet dense granules, and is called inorganic polyphosphate”, explains Dr. Felix A. Ruiz, head of the research team of the University and Hospital Puerta del Mar.

In some patients, dense granules of platelets are deficient in number and function, an anomaly called "Disease of the Delta Reservoir" or "Delta-Storage Pool Disease" (DSPD) in English. The disease is manifested by a moderate tendency to bleed, but sometimes, as in surgery or after childbirth, heavy bleeding may occur and it can endanger the patient's life.

So far, the diagnosis of the disease is very complicated with tests that require expensive equipment and laborious procedures. The new proposal creates a rapid diagnostic kit, simple and affordable for any health centre.

“It is believed that this disease is not common among the population, but the experts think that many cases have not been diagnosed yet”. Due to the complex procedure that currently exists for the diagnosis, “only studied patients who have had serious problems with bleeding or very repetitive”, argues Dr. Ruiz.

The Delta Reservoir disease is usually caused by a congenital defect but may develop as a result of other diseases such as leukaemia or liver disease. According to Dr. Ruiz says, “The use of the procedure presented, would determine the actual incidence of the disease in the general population and routinely examine patients who are at risk of developing it”.

To carry out the procedure it has been necessary to conduct a study in which 130 healthy individuals have collaborated, who provided samples were compared with those of a group of patients with the disease. Besides generating patents, the results of this study have been published in the Journal of Thrombosis and Haemostasis, one of the ten best magazines of Haematology of the world.

This invention develops kits for evaluation and testing of polyphosphate levels in both isolated platelets and in other cell types. “It is appropriate to examine a large number of samples in a short time, and additionally also be used to diagnose the disease in other mammals, some of commercial importance, as in some pedigree dogs and cattle where the highest incidence were found”, says Felix A. Ruiz.

The inventors of this technique are Laura Hernández Ruiz, Ana Saez Benito, Maria D. Núñez Jiménez, Javier Rodríguez Martorell and Felix A. Ruiz, who mostly are working around the polyphosphate, “Our new hypothesis is based on other platelet defects, which may also be reflected in changes in polyphosphate”. Since the investigators have already the standards of healthy people, the experts have checked the defect of dense granules, now the researchers will examine samples from individuals with other abnormalities in their platelets. “We are able to begin to see if any platelet related disease is reflected in a variation of polyphosphate and if so, could lead to new patents and technological developments”, says the doctor.
